Abstract

Abstract: With the recent excitement around the cancer moonshot initiative, one might naturally ask: what would the 1969 moonshot have looked like if we engineered space travel the same way we develop oncology drugs today? (Spoiler alert: not good.) We then contrast the current oncology development paradigm with a proposed 'rocket science' approach using continuous time-dependent measurements and predictive models to optimize probability of success for investigational drugs and ultimately cancer patients. We will show several examples of this approach, touching on modeling methods for safety and efficacy evaluation of novel-novel drug combinations, quantification of the effect of baseline biomarkers on tumor kinetics, and characterization of dose and schedule-response properties of an investigational immuno-oncology agent.
